This work concentrates on cellular and molecular toxicity of selected well-known drugs or chemicals on the cardiovascular system. The primary objective is to provide a better understanding of the mechanisms by which xenobiotics are toxic to mammalian tissues and cells. The use of in vitro cellular and tissue systems provides attractive experimental models to assess toxic manifestations of xenobiotics. This work addresses the most recent findings on the cellular and molecular mechanisms of toxicity of several important cardiotoxic agents: doxorubicin, ethanol, cocaine, and the catecholamines. It presents an overview of vascular toxins and their biochemical effects. Included is a summary of in vitro cardiovascular techniques for assessing toxicity of xenobiotics. This publication is important for those in toxicology, tissue culture, pharmacology, in vitro toxicology, developmental biology and related areas.

Advances in Molecular Toxicology features the latest advances in all of the subspecialties of the broad area of molecular toxicology. Toxicology is the study of poisons, and this series details the study of the molecular basis by which a vast array of agents encountered in the human environment and produced by the human body itself manifest themselves as toxins. Not strictly limited to documenting these examples, the series is also concerned with the complex web of chemical and biological events that give rise to toxin-induced symptoms and disease. The new technologies that are being harnessed to analyze and understand these events will also be reviewed by leading workers in the field. Advances in Molecular Toxicology will report progress in all aspects of these rapidly evolving molecular aspects of toxicology with a view toward detailed elucidation of progress on the molecular level and on advances in technological approaches employed. Cutting-edge reviews by leading workers in the discipline In-depth dissection of molecular aspects of interest to a broad range of scientists, physicians and any student in the allied disciplines Leading edge applications of technological innovations in chemistry, biochemistry and molecular medicine

Volume 14 in the series Comprehensive Toxicology extends and complements the previously published 13-volume set. This volume will be available separately. Toxicology is the study of the nature and actions of chemicals on biological systems. In more primitive times, it really was the study of poisons. However, in the early 1500s, it was apparent to Paracelsus that "the dose differentiates a poison and a remedy". Clearly, the two most important tenets of toxicology were established during that time. The level of exposure (dose) and the duration of exposure (time) will determine the degree and nature of a toxicological response. Since that time the discipline of toxicology has made major advances in identifying and characterizing toxicants. The growth of toxicology as a scientific discipline has been driven to a large extent by the use of extremely powerful molecular and cell biology techniques. The overall aim of this volume is to demonstrate how these advances are being used to elucidate causal pathways (or linkages) for potential adverse health consequences of human exposure to environmental chemicals or radiation. A unique feature of this volume is its illustration of how carefully-designed studies of the molecular mechanisms of chemical action provide not only understanding of the potential toxicity of the chemical under investigation, but also new insights into the functioning of the biological system used as an experimental model. Each chapter contains a listing of major peer-reviewed articles and reviews and useful web-sites. In addition, each chapter contains a broad introductory section that outlines the subsequent sections. These Introductory and Overview sections are designed to be stand alone chapters, and may be packaged as a textbook in graduate level courses.

In Vitro Toxicology details the protocols and methods of in vitro testing, highlighting the usefulness of models, methods and the cost-effectiveness and reproducibility of such methodologies. The current approaches and strategies required to develop an easy, reliable, validated and high throughput system for use in alternative animal models to circumvent in vivo testing are discussed in detail. The book also includes chapters on the principles involved in the general selection and use of models that address safety concerns, regulatory acceptance and the current understandings and strategies for the identification of biomarkers in toxicity testing. Furthermore, principles involved in the general selection and use of models that address the issues of safety concerns and regulatory acceptance of these models are discussed, making the book beneficial to students, scientists, and regulators working in toxicology, as well as those in the field of chemicals and the safety assessment of novel materials. Discusses new techniques and protocols in a clear and concise manner includes examinations of nanotoxicity, genotoxicity and carcinogenicity Explains practical laboratory methods and the theories behind in vitro testing

In response to the concerns voiced by Vietnam veterans and their families, Congress called upon the National Academy of Sciences (NAS) to review the scientific evidence on the possible health effects of exposure to Agent Orange and other herbicides. This call resulted in the creation of the first NAS Institute of Medicine Committee to Review the Health Effects in Vietnam Veterans of Exposure to Herbicides in 1992. The committee published its initial findings in the 1994 report Veterans and Agent Orange: Health Effects of Herbicides Used in Vietnam. This report is the result of a 1999 request from the Department of Veterans Affairs (DVA) under the aegis of the Veterans and Agent Orange research program. Specifically, DVA asked the committee to examine evidence regarding the association, if any, between Type 2 diabetes and exposure to dioxin and other chemical compounds in herbicides used in Vietnam. Veterans and Agent Orange: Herbicide/Dioxin Exposure and Type 2 Diabetes reviews the scientific evidence regarding the association, if any, between Type 2 diabetes1 and exposure to dioxin2 and other chemical compounds in herbicides used in Vietnam. This report examines, to the extent that available data permitted meaningful determinations, (1) whether a statistical association with herbicide exposure exists, taking into account the strength of the scientific evidence and the appropriateness of the statistical and epidemiologic methods used to detect the association; (2) the increased risk of the disease among those exposed to herbicides during Vietnam service; and (3) whether there is a plausible biological mechanism or other evidence of a causal relationship between herbicide exposure and the disease.

The pollution of the earth's freshwater habitats is a topic of major concern. This timely synthesis considers the effects of pollutants on aquatic animals via a series of research and review articles which present experimental evidence of sublethal and lethal effects of a range of toxicants at the physiological, cellular and subcellular levels. Topics covered include routes of uptake of toxicants; the effects of acute and chronic exposure to toxic metal ions, particularly zinc, copper and aluminium, with emphasis on the mechanisms of toxicity and chronic exposure to sublethal levels; the impact on fish biology of two chemicals of current concern, nitrites and oestrogenic substances; and studies of the mechanisms of toxicity at the cellular and subcellular level, including damage of DNA, using cultured fish cells.
